I received an American Express gift card worth $150. I listed it in my account to be used for purchases. If I set that as the "preferred" card, and then make a purchase for more than $150, will the balance of the payment automatically shift over to the credit card that I commonly use?

No, the card would decline. The card must have enough on it to cover purchase.
