I'm looking to sell a gift card on ebay as either a digital item or posted item, which method has the most protection against fraudulent claims? I've sold Xbox Live gift cards in the past where someone has reported the transaction for whatever reason and got their money back (then I've lost the gift card) and I really don't want to get stung again, especially as the value of this one is £400.

Read the 11. Seller Protection Programme
Mail it with a tracking number to get seller protection but at the same time gift cards are not covered in general. You may have a fighting chance if you physically mailed it though, rather than sending it via email.
Also watch out for scams because there are other ways to skin a cat:

It sound really stupid but what would your gut instinct tell you to do?
At that amount, I can ill afford to be burned by buyers who would inevitably chance their luck.
The risk seems higher than the reward currently.

I would see if a good friend or family member or signficant other has a birthday coming up that you can gift the card to. Or consider donating the card to a charity and write it off as a tax write-off.
